What's your message?
Think of the message you want to get across. Conciliatory, confident, stress your strengths.
Timing!
Make it no longer than 10 minutes - don't waffle, keep your points concise.
Look into the whites of their eyes!
Remember you are not addressing the mediator like you would a Judge - he has read the papers, he probably understands the facts, so this gives you an opportunity to speak directly to the other side and their client across the table. Make the most of it!
Weak case?
Tell them (or their client) the weaknesses in their case - politely, of course!
Ending 
Finish on a conciliatory note.
